    Management and training of linguistic volunteers: a case study of translation at Cochrane Germany

    Cochrane is a global, non-profit organisation that synthesizes health-related research evidence. It established a translation strategy in 2014 to increase the significance of its information beyond the English-speaking world. Under the strategy, translation at Cochrane is achieved mostly through the efforts of linguistic volunteers. Translation in crisis settings, too, relies on the work of volunteers; however, appropriate ways to manage and train these volunteers are unclear. We carried out a study of the case of translation at one part of Cochrane, Cochrane Germany, to learn about the management and training of linguistic volunteers there and in Cochrane more broadly. Thematic analysis of data gathered by the researcher during a two-month secondment to the offices of Cochrane Germany– including data from formal interviews, informal meetings, field notes, a reflective journal, and a large corpus of grey literature – generated three main themes. The themes relate to appropriate conceptualisations of linguistic volunteers, project management in the assurance of quality volunteer work, and feedback as a form of volunteer training. Recommendations are made to apply these lessons learned to future work on crisis translation and for possible improvements to linguistic volunteer management and training at Cochrane

    Non-target screening with high-resolution mass spectrometry: critical review using a collaborative trial on water analysis

    In this article, a dataset from a collaborative nontarget screening trial organised by the NORMAN Association is used to review the state-of-the-art and discuss future perspectives of non-target screening using high-resolution mass spectrometry in water analysis. A total of 18 institutes from 12 European countries analysed an extract of the same water sample collected from the River Danube with either one or both of liquid and gas chromatography coupled with mass spectrometry detection. This article focuses mainly on the use of high resolution screening techniques with target, suspect, and non-target workflows to identify substances in environmental samples. Specific examples are given to emphasise major challenges including isobaric and co-eluting substances, dependence on target and suspect lists, formula assignment, the use of retention information, and the confidence of identification. Approaches and methods applicable to unit resolution data are also discussed. Although most substances were identified using high resolution data with target and suspect-screening approaches, some participants proposed tentative non-target identifications. This comprehensive dataset revealed that nontarget analytical techniques are already substantially harmonised between the participants, but the data processing remains time-consuming. Although the objective of a Bfullyautomated identification workflow^ remains elusive in the short term, important steps in this direction have been taken, exemplified by the growing popularity of suspect screening approaches. Major recommendations to improve non-target screening include better integration and connection of desired features into software packages, the exchange of target and suspect lists, and the contribution of more spectra from standard substances into (openly accessible) databases.This work was supported in part by the SOLUTIONS project, which received funding from the European Union’s Seventh Framework Programme for research, technological development and demonstration under Grant Agreement No. 603437

    Inter-laboratory mass spectrometry dataset based on passive sampling of drinking water for non-target analysis

    Non-target analysis (NTA) employing high-resolution mass spectrometry is a commonly applied approach for the detection of novel chemicals of emerging concern in complex environmental samples. NTA typically results in large and information-rich datasets that require computer aided (ideally automated) strategies for their processing and interpretation. Such strategies do however raise the challenge of reproducibility between and within different processing workflows. An effective strategy to mitigate such problems is the implementation of inter-laboratory studies (ILS) with the aim to evaluate different workflows and agree on harmonized/standardized quality control procedures. Here we present the data generated during such an ILS. This study was organized through the Norman Network and included 21 participants from 11 countries. A set of samples based on the passive sampling of drinking water pre and post treatment was shipped to all the participating laboratories for analysis, using one pre-defined method and one locally (i.e. in-house) developed method. The data generated represents a valuable resource (i.e. benchmark) for future developments of algorithms and workflows for NTA experiments

    Full-Length L1CAM and Not Its Δ2Δ27 Splice Variant Promotes Metastasis through Induction of Gelatinase Expression

    Tumour-specific splicing is known to contribute to cancer progression. In the case of the L1 cell adhesion molecule (L1CAM), which is expressed in many human tumours and often linked to bad prognosis, alternative splicing results in a full-length form (FL-L1CAM) and a splice variant lacking exons 2 and 27 (SV-L1CAM). It has not been elucidated so far whether SV-L1CAM, classically considered as tumour-associated, or whether FL-L1CAM is the metastasis-promoting isoform. Here, we show that both variants were expressed in human ovarian carcinoma and that exposure of tumour cells to pro-metastatic factors led to an exclusive increase of FL-L1CAM expression. Selective overexpression of one isoform in different tumour cells revealed that only FL-L1CAM promoted experimental lung and/or liver metastasis in mice. In addition, metastasis formation upon up-regulation of FL-L1CAM correlated with increased invasive potential and elevated Matrix metalloproteinase (MMP)-2 and -9 expression and activity in vitro as well as enhanced gelatinolytic activity in vivo. In conclusion, we identified FL-L1CAM as the metastasis-promoting isoform, thereby exemplifying that high expression of a so-called tumour-associated variant, here SV-L1CAM, is not per se equivalent to a decisive role of this isoform in tumour progression

    Effect of angiotensin-converting enzyme inhibitor and angiotensin receptor blocker initiation on organ support-free days in patients hospitalized with COVID-19

    IMPORTANCE Overactivation of the renin-angiotensin system (RAS) may contribute to poor clinical outcomes in patients with COVID-19. Objective To determine whether angiotensin-converting enzyme (ACE) inhibitor or angiotensin receptor blocker (ARB) initiation improves outcomes in patients hospitalized for COVID-19. DESIGN, SETTING, AND PARTICIPANTS In an ongoing, adaptive platform randomized clinical trial, 721 critically ill and 58 non–critically ill hospitalized adults were randomized to receive an RAS inhibitor or control between March 16, 2021, and February 25, 2022, at 69 sites in 7 countries (final follow-up on June 1, 2022). INTERVENTIONS Patients were randomized to receive open-label initiation of an ACE inhibitor (n = 257), ARB (n = 248), ARB in combination with DMX-200 (a chemokine receptor-2 inhibitor; n = 10), or no RAS inhibitor (control; n = 264) for up to 10 days. MAIN OUTCOMES AND MEASURES The primary outcome was organ support–free days, a composite of hospital survival and days alive without cardiovascular or respiratory organ support through 21 days. The primary analysis was a bayesian cumulative logistic model. Odds ratios (ORs) greater than 1 represent improved outcomes. RESULTS On February 25, 2022, enrollment was discontinued due to safety concerns. Among 679 critically ill patients with available primary outcome data, the median age was 56 years and 239 participants (35.2%) were women. Median (IQR) organ support–free days among critically ill patients was 10 (–1 to 16) in the ACE inhibitor group (n = 231), 8 (–1 to 17) in the ARB group (n = 217), and 12 (0 to 17) in the control group (n = 231) (median adjusted odds ratios of 0.77 [95% bayesian credible interval, 0.58-1.06] for improvement for ACE inhibitor and 0.76 [95% credible interval, 0.56-1.05] for ARB compared with control). The posterior probabilities that ACE inhibitors and ARBs worsened organ support–free days compared with control were 94.9% and 95.4%, respectively. Hospital survival occurred in 166 of 231 critically ill participants (71.9%) in the ACE inhibitor group, 152 of 217 (70.0%) in the ARB group, and 182 of 231 (78.8%) in the control group (posterior probabilities that ACE inhibitor and ARB worsened hospital survival compared with control were 95.3% and 98.1%, respectively). CONCLUSIONS AND RELEVANCE In this trial, among critically ill adults with COVID-19, initiation of an ACE inhibitor or ARB did not improve, and likely worsened, clinical outcomes.     An account on BiVO4 as photocatalytic active matter

    Due to their controllability, photocatalytic materials are gaining in popularity and research investment. While most of the early work used highly stable TiO2 as a material to construct micromotors, mostly in combination with noble metals, other semiconductors offer a wider range of properties, including independence from high-energy UV light. This review focuses on our work with BiVO4, which has shown promise due to its smaller band gap which absorbs blue light, and its well-defined crystal structures that can lead to charge separation on different areas of the crystal, providing sufficient asymmetry to cause active propulsion. These properties have given rise to fascinating physical and chemical behaviours that show how rich and variable active matter can become. This review summarises our early findings and relates them to the inherent material properties and characteristics of BiVO4. We present the synthesis of different microparticles and their material properties that make them excellent candidates as active micromotors. A critical factor in understanding inherently asymmetric micromotors is knowledge of their flow fields. However, due to the small size of the particles and the need to use even smaller tracer particles to avoid perturbing the flow field, measuring flow fields at the microscale is a difficult task. We also present these first results, which allow us to demonstrate the correlation between chemical reactivity and the flow generated, leading to active motion. Due to the non-toxic nature of BiVO4, these novel microswimmers have been used to study the first steps towards applications, even in sensitive areas such as food technology. Although these initial tests are far from being realised, we have to face the fact that a single microswimmer will not be able to perform macroscale tasks. Therefore, we present the reader with the first simple studies of collective motion, hoping for many new contributions to the field. The one-step synthesis of BiVO4 clearly paves the way for studies requiring large numbers of particles. We predict that the combination of promising applications for a non-toxic material which is readily synthesised in large quantities will contribute pivotally to develop the field of active matter beyond the proof-of-concept stage
